1. Basically you want the current mods to not feel there's something "fishy" or "suspect" about you. How you accomplish that is up to you. Letting the mods get to know you is usually a good way of doing this.
2. Then you need to show you're competent. Don't make complaint threads or file reports which show you have a poor understanding of the rules. Comment on threads and reports in such a way that shows you have an understanding of the rules and how they're moderated.
3. Don't insult people. Chances are at least 1 person on the mod team will take offense to what you said and consider you a mean person for it and tell other mods you're a mean person.
4. Don't try too hard or for some reason you don't get picked. Probably relates to causing job (1.) to fail.

You can apply over and over for like 3 years and people will be modded that have already had the job before again and again. It's not worth it for the amount of time it would actually take. I suggest making friends with the right people.
Don't cheat, be on the site for a long time and most of all infiltrate yourself into the friend-zone of current moderators or just simply boot-lick the admin.
